{"id":26982,"date":"2024-01-02T05:00:18","date_gmt":"2024-01-02T10:00:18","guid":{"rendered":"https:\/\/orionscoldfire.com\/?p=26982"},"modified":"2024-01-02T15:57:11","modified_gmt":"2024-01-02T20:57:11","slug":"tolerance-proteins","status":"publish","type":"post","link":"https:\/\/orionscoldfire.com\/index.php\/2024\/01\/02\/tolerance-proteins\/","title":{"rendered":"Tolerance Proteins"},"content":{"rendered":"<p>In 2024 I\u2019ve resolved to do my best not to belabor things that have already been said.\u00a0 Therefore, going forward, I will attempt to confine myself to original content.\u00a0 So, what can I talk about?\u00a0 Breaking news?\u00a0 Well, there was a major earthquake in Japan.<\/p>\n<p>But there\u2019s always a major earthquake in Japan.\u00a0 Hauling out terms like temblor and Richter scale and seismic this, that and the other or going on and on about aftershocks and tsunamis is hardly original.\u00a0 Pass!<\/p>\n<p>Obviously, the wars in Gaza and Ukraine are just grinding along in a predictably gruesome fashion.\u00a0 Nope.<\/p>\n<p>Well, I could conceive of nothing less original than the ball-dropping ceremony in Times Square and its attendant gathering of aging and uninteresting entertainment celebrities.\u00a0 Chah!<\/p>\n<p>So, no breaking news worth discussing.<\/p>\n<p>How about tardigrades?<\/p>\n<p><iframe loading=\"lazy\" title=\"The INVINCIBLE Animal!\" width=\"783\" height=\"440\" src=\"https:\/\/www.youtube.com\/embed\/hbNFfbp7YGA?feature=oembed\" frameborder=\"0\" allow=\"accelerometer; autoplay; clipboard-write; encrypted-media; gyroscope; picture-in-picture; web-share\" allowfullscreen><\/iframe><\/p>\n<p>You know, water bears.\u00a0 Those barely visible invertebrates that can be exposed to the hard vacuum and near absolute zero of outer space, to hard radiation, or desiccated in a desert for years and even frozen in ice and boiled in geysers and still wake up decades later and resume feeding on moss or dandruff or whatever is available and seem completely unphased.<\/p>\n<p>Finally, something interesting to talk about.<\/p>\n<p>Ever since I first heard about these bizarre and strangely endearing little creatures, I\u2019ve wondered whether there was something unique about their survival abilities.\u00a0 And it turns out there is.<\/p>\n<p>\u201c<a href=\"https:\/\/journals.plos.org\/plosbiology\/article?id=10.1371%2Fjournal.pbio.3001780\" target=\"_blank\" rel=\"noopener\">Stress-dependent cell stiffening by tardigrade tolerance proteins that reversibly form a filamentous network and gel<\/a>\u201d<\/p>\n<p>As you know I\u2019m no longer a big fan of tolerance.\u00a0 It\u2019s bitten me in the butt too many times over the years.\u00a0 But apparently <strong>tolerance proteins<\/strong> are a very useful thing to have when you find yourself submerged in freezing liquid or exposed to outer space.<\/p>\n<p><strong>\u201cTardigrades are able to tolerate almost complete dehydration by entering a reversible ametabolic state called anhydrobiosis and resume their animation upon rehydration. Dehydrated tardigrades are exceptionally stable and withstand various physical extremes.\u201d<\/strong><\/p>\n<p>\u2026<\/p>\n<p>\u201c<strong>In the present study, we first postulated the presence of tolerance proteins that form protective condensates via phase separation in a stress-dependent manner and searched for tardigrade proteins that reversibly form condensates upon dehydration-like stress.\u201d<\/strong><\/p>\n<p><strong>\u2026<\/strong><\/p>\n<p><strong>CAHS proteins increased cell stiffness in a hyperosmotic stress-dependent manner and counteract the cell shrinkage caused by osmotic pressure, and even improved the survival against hyperosmotic stress.<\/strong><\/p>\n<p><strong>\u2026<\/strong><\/p>\n<p><strong>On the basis of these results, we propose that CAHS proteins are novel cytoskeleton-like proteins that form filamentous networks and undergo gel-transition in a stress-dependent manner to provide on-demand physical stabilization of cell integrity against deformative forces during dehydration and could contribute to the exceptional physical stability in a dehydrated state.<\/strong><\/p>\n<p>I <a href=\"https:\/\/youtu.be\/0vc0TTQZEcU?t=685\" target=\"_blank\" rel=\"noopener\">heard about a Chinese study<\/a> that inserted genes from a tardigrade into a human cell line that gave it protection from radiation associated DNA damage.\u00a0 It was posited that this would be a valuable addition to the creation of human super soldiers.\u00a0 Well duh!\u00a0 Anyone who has watched Captain America knows that tardigrade-style ability to tolerate the radiation from a tesseract plus the ability to be frozen for a century or so in the Arctic Ocean is very handy for a super soldier.<\/p>\n<p>My takeaway from all this is that adding tardigrade tolerance proteins will allow humans to be placed in a long term reversible ametabolic state called anhydrobiosis from which they will be able to emerge after millennia in order to serve as my laborers once I emerge from long sleep in some far away galaxy.\u00a0 As their emperor I will be able to call on their immunity to radiation to have them wage nuclear war against my enemies the beetle men of Krrrrlll 3,<\/p>\n<p><iframe loading=\"lazy\" title=\"1967 Atom Cartoon -
